چکیده

The specific role of neuroinflammation in the pathogenesis of Parkinson's disease remains to be fully elucidated. By infusing lipopolysaccharide (LPS) into the striatum, we investigated the effect of neuroinflammation on the dopamine nigrostriatal pathway. Here, we report that LPS-induced neuroinflammation in the striatum causes progressive degeneration of the dopamine nigrostriatal system, which is accompanied by motor impairments resembling parkinsonism. Our results indicate that neurodegeneration is associated with defects in the mitochondrial respiratory chain related to extensive S-nitrosylation/nitration of mitochondrial proteins. Mitochondrial injury was prevented by treatment of L-N 6-(l-iminoethyl)-lysine, an inducible nitric oxide synthase (iNOS) inhibitor, suggesting that iNOS-derived NO is responsible for mitochondrial dysfunction. Furthermore, the nigral dopamine neurons exhibited intracytoplasmic α-synuclein and ubiquitin accumulation. These results demonstrate that degeneration of nigral dopamine neurons by neuroinflammation is associated with mitochondrial malfunction induced by NO-mediated S-nitrosylation/nitration of mitochondrial proteins.

B Vitamins Supplement Potentiates Antiparkinsonian Effect of Flunarizine: the Behavioral and Biochemical Evidences From 6-Hydroxydopamine Animal Model

Introduction: Prominent data indicate that flunarizine (flu), a calcium channel blocker, has neuroprotective effect. However, several authors have reported that the chronic use of flu can produce drug-induced Parkinsonism. Previously, we showed that B vitamins supplement (B com) has antiparkinsonian effect. In the present study, we evaluated the effect of pretreatment with flu ...

P 12: Study of the Association between Serum Level of Cystatin C and Behavioral Symptoms of 6-Hydroxydopamine – Induced Parkinsonism in Rat

Introduction: Parkinson’s disease (PD) is the second most neurodegenerative disorder which is characterized by a progressive loss of dopaminergic neurons in the substantia nigra pars compacta. Clinical symptoms do not appear until approximately 70% of dopaminergic neurons and 80% of the striatal dopaminergic terminals have been lost. Thus, detecting nonclinical factors such as detecting b...

Striatal Neuroinflammation Promotes Parkinsonism in Rats

BACKGROUND Sporadic Parkinson's disease (PD) is a progressive neurodegenerative disorder with unknown cause, but it has been suggested that neuroinflammation may play a role in pathogenesis of the disease. Neuroinflammatory component in process of PD neurodegeneration was proposed by postmortem, epidemiological and animal model studies. However, it remains unclear how neuroinflammatory factors ...

Determination of the Role of Synapse Associated Protein 97 (SAP97) in the Normal and Parkinsonian Striatum By

Parkinson’s disease (PD) is a debilitating movement disorder associated with the death of dopaminergic nigrostriatal neurons. In addition to dopamine deficiency, abnormalities in glutamate and other receptors at striatal synapses have been reported. Synapse associated protein 97 (SAP97) is involved in regulation of glutamate receptor function. In the striatum of unilaterally-lesioned 6-OHDA rat...

Extra-striatal Uptake of 99mTc-TRODAT SPECT in a Cerebral Meningioma: A Case Report

We reported a 71 years old woman, with history of rest and postural tremor, bradykinesia and memory problems. In her dynamic MRI, a contrastenhanced tumor in the cerebellopontine (CP) angle was found which was compatible with a meningioma. 99mTc-TRODAT SPECT showed decreased activity in the left putamen, indicating idiopathic Parkinson disease. There was also a focus of increased activity on th...
